Sildenafil and its pharmacologic effects in the treatment of erectile dysfunction

Sildenafil is a phosphodiesterase-5 inhibitor (PDE5-5 inhibitor). It was developed by Eli Lilly and Company in 2003 and approved by the U. S. Food and Drug Administration in 2010.

Sildenafil is also approved by the European Medicines Agency for the treatment of erectile dysfunction (ED).

How it works

Sildenafil and its active ingredient, the sildenafil citrate, work together to inhibit the enzyme phosphodiesterase type 5 (PDE5). This enzyme breaks down cGMP, the substance that makes penile blood vessels dilate and relax, causing an erection to occur. By inhibiting PDE5, sildenafil enhances the effects of PDE5, allowing the smooth muscles to relax and widen.

PDE5 is the chemical name for a group of chemicals that are called phosphodiesterase-5 inhibitors. Sildenafil is the active ingredient in Viagra. It works by relaxing the smooth muscles of the penis and allowing blood to flow into the penis when sexually aroused.

Sildenafil is a short-acting selective PDE5 inhibitor, which means it doesn’t inhibit the enzyme PDE5. It works by dilating the blood vessels in the penis, allowing the blood to flow into the penis when sexually excited.

Sildenafil, like Viagra, is prescribed in combination with other medications for erectile dysfunction, such as tadalafil. It’s also sometimes prescribed in combination with other medications for pulmonary arterial hypertension, where it works by relaxing the blood vessels, helping the heart work properly.

How sildenafil works
